Flow cytometry using fluorescein (FITC) and phycoerythrin (PE) or other fluorochrome-conjugated monoclonal antibodies against CD19 and CD103 surface antigens, followed by acquisition on a clinical flow cytometer and absolute count calculation using counting beads or dual-platform methodology with concurrent complete blood count (CBC) with differential.

Specimen should be processed within 24 to 48 hours of collection when stored at room temperature (18–25 °C). Prolonged storage beyond 48 hours may result in loss of cell viability and altered surface antigen expression. Do not refrigerate or freeze whole blood specimens intended for flow cytometry.

Reference range

Reference ranges for CD19+CD103+ co-expressing B lymphocytes in peripheral blood are not well-established in standard clinical literature. CD19+CD103+ cells represent a rare B-cell subset, typically comprising a very small fraction of circulating B lymphocytes. Values are laboratory-specific; results should be interpreted using the performing laboratory's established reference interval. Studies in healthy adults generally report very low or undetectable levels of CD19+CD103+ cells in peripheral blood.N/A

No special patient preparation is required. Fasting is not necessary. Patients should be well-hydrated. Recent corticosteroid therapy, chemotherapy, immunotherapy, or immunosuppressive medications may affect lymphocyte subset distribution and should be noted on the requisition.

Clinical interpretation

How abnormal results are interpreted and what conditions they suggest.
